Biotransformation of Ginsenoside Rb1 to Ginsenoside Rd by ¥â-glucosidase from Lactobacillus plantarum G1

Abstract
The ¥â-glucosidase from Lactobacillus plantarum G1, isolated from Kimchi, was purified as a single 50 kDa band by ammonium sulfate precipitation, HiTrap Q Hp, and Resource Q column chromatography, with a specific activity of 37 U/mg. The ¥â- glucosidase L. plantarum G1 showed optimum activity at pH 4.0 and 40¡É for ginsenoside Rd production. The ¥â-glucosidase from L. plantarum G1 hydrolyze only outer ¥â-(1,2)-D-glucose linkage at C-20 position in ginsenosides which has narrow substrate specificity for ginsenoside. Under the optimum conditins, the enzyme produced 1 mM of ginsenoside Rd from 1 mM of ginsenoside Rb1 after 120 min, with molar yield of 100%.
